The method which I have imployed is as follows:-
The frogs are Killed by Chloroform and pinned on a flat board. With sterilised instruments the heart is laid bare. The surface of the latter is sterilised by a hot spatula or other convenient instrument, and then by sterilised Pasteur's pipette the contained blood is withdrawn and immediately transferred to the culture medium. If this is done with an ordinary amount of careful technique, little difficulty will be found in obtaining pure cultures, Cultures free from ordinary micro-organisms. As already mentioned, the cytamabae bactifera show little tendency to multiply in ordinary culture media.
